Accurate identification methods will play an important role in disease control management. Flagellum is the main commodity of the bacteria that facilitates its movement. Furthermore, it is of particular importance in colonization and pathogenicity. The fliC gene is one among five other genes encoding flagellin, the protein that makes the flagella in bacteria. In this study, the flagellin genes of 11 bacterial species were selected and the phylogenetic tree analysis divided them into plant and animal bacteria groups. Electron microscope images were taken from the bacterial flagella and its length was up to nine microns with 400-600 nm in diameter; as observed in both Lophotrichous and Monotrichous forms. fliC was amplified and analyzed for the first time by polymerase chain reaction for the two isolates of bacterial pathogen of saffron. Specific primers were designed and used that managed to separate Bgg from other bacteria, and helped in distinguishing the infected saffron corms from the healthy. Electron microscopy of the flagellum was illustrative of different types of Bgg. Investigating and studying the fliC gene can be useful in diagnosis of the causative agent of saffron corm rot disease.}, keywords = {Burkholdria gladioli",Electron Microscopy,flagella",flagellin",",phylogenetic tree"}, title_fa = {مطالعه تاژک و ژن fliC باکتری عامل بیماری پوسیدگی بنه زعفرانBurkholderia gladioli pv. gladioli، راهبردی در شناسایی باکتری و پیشگیری از گسترش بیماری}, abstract_fa = {باکتری Burkholderia gladioli pv. gladioli (Bgg) عامل بیماری پوسیدگی بنه زعفران از بیماریهای شایع این گیاه است. روشهای شناسایی دقیق نقش مهمی در مدیریت کنترل بیماری خواهند داشت. تاژک، وسیله اصلی حرکت در باکتریها است و از اهمیت خاصی در کلونیزاسیون، استقرار و بیماریزایی برخوردار است. ژن fliC یکی از پنج ژن کد کننده فلاژلین میباشد. در این مطالعه، ژنهای فلاژلین یازده گونه باکتریایی انتخاب شدند که با آنالیز فیلوژنتیکی در گروههای باکتریهای گیاهی و جانوری جای گرفتند. تصاویر میکروسکوپ الکترونی از تاژک طول آن را حداکثر 9 میکرون با قطری معادل 600-400 نانومتر به دو شکل لوفوتریش و مونوتریش نشان داد. ژن fliC  برای اولین بار به کمک PCR در دو جدایه باکتری جدا شده از گیاه زعفران تکثیر و مورد بررسی قرار گرفت. پرایمرهای اختصاصی طراحی شده بطور مؤثری توانستند این باکتری را از سایر گونههای دیگر تفکیک نمایند و به سرعت بنههای آلودهی زعفران را از بنههای سالم تشخیص دهند. با استفاده از تصاویر میکروسکوپ الکترونی، تاژک از منظر نوع آرایش در دو جدایه باکتری جدا شده از گیاه زعفران مورد بررسی قرار گرفت. بررسی و مطالعه ژن fliC میتواند در تشخیص عامل بیماری پوسیدگی بنه زعفران مفید واقع شود.}, keywords_fa = {بورخولدریا گلادیولی,تاژک,درخت فیلوژنتیکی,فلاژلین,میکروسکوپ الکترونی}, url = {https://saffron.torbath.ac.ir/article_159137.html}, eprint = {https://saffron.torbath.ac.ir/article_159137_d30b6a2edbf520d42ab64fd9b7be24a3.pdf} }
